Protein Chemistry: Casein, Denaturation, and Coagulation

Casein is a group of phosphoproteins found in mammalian milk, making up about 80% of the proteins in cow's milk. It is a key component in cheese production and has various other uses, including as an emulsifier and in adhesives. Understanding casein is important because it plays a central role in food science and technology. Denaturation is the process by which proteins lose their native structure due to external factors like heat, pH changes, or chemical agents. This structural change can lead to a loss of function, such as an enzyme no longer being able to catalyze reactions. Coagulation, a result of denaturation, is the process where proteins lose solubility and form a solid mass. This is evident in everyday examples like the solidification of egg whites when cooked or the curdling of milk to make cheese. It makes up most of the protein in cow's milk and helps give milk its structure. Casein proteins are held together in tiny clusters called micelles, which keep them from clumping in normal milk. Denaturation is when a protein loses its usual shape.

This can happen from heat, strong acid or base, or other chemicals. When a protein is denatured, it often becomes less useful or stops working. For example, enzymes can no longer speed up reactions if they are denatured.

When casein is denatured, it becomes less soluble and starts to form solid clumps. This is why milk curdles when you add acid or heat. In cheese-making, this process turns liquid milk into solid curds.

The same thing happens when you cook egg whites and they turn from clear liquid to firm white solid. When something changes the environment, like adding acid or heat, these balls unfold and stick together, forming a solid mass. This is how cheese, glue, and even the cooked parts of eggs are made.
